We appreciate Dr Li’s enthusiasm of the study findings as well as the well-thought-out critique of the manuscript.1 Transcription and replication are essential processes in normal and cancer cells alike. Expectedly, therapeutic targeting of each of these processes alone results in toxicity. Based on the data presented in our paper, we identified transcription-replication conflicts (TRCs) as a maladaptive response that is much more common in cancer cells caused by oncogene-driven dysregulation of both transcription and replication.
